Evolution of Human Phosphorylation Sites
نویسندگان
چکیده
Phosphorylation of proteins plays key roles in many intracellular signal pathways [3]. Gain or loss of phosphorylation sites of proteins during evolution may result in changes in signal transduction pathways. For example, FOXP2, a gene involved in speech and language, was reported to gain a potential target site for phosphorylation in humans after the human-chimpanzee divergence [2]. Here, we report development and application of a semi-automatic method for identification of novel phosphorylation sites that are newly acquired in human proteins.

Turnover of protein phosphorylation evolving under stabilizing selection
Most proteins are regulated by posttranslational modifications and changes in these modifications contribute to evolutionary changes as well as to human diseases. Phosphorylation of serines, threonines, and tyrosines are the most common modifications identified to date in eukaryotic proteomes. While the mode of action and the function of most phosphorylation sites remain unknown, functional stu...
متن کاملO-7: Insulin Exerts a Prosurvival Effect on Human Spermatozoa via Mechanisms That Involve the Stimulation of Akt Phosphorylation
Background: The purpose of this study was to examine the impact of Insulin (INS) on human sperm function, in light of a recent proteomic analysis indicating that these cells express the INS receptor (INSR). Materials and Methods: We used Western blot and Immunocytochemical analyses. Results: Immunocytochemical analyses confirmed the presence of INSR in human spermatozoa and localized this recep...

The aim of the present study was todeterminecharacteristics of Rheb gene and protein in Raini Cashmere goat. Comparative analyses of the nucleotide sequences were performed. Open reading frames (ORFs), theoretical molecular weights of deduced polypeptides, the protein isoelectric point, protein characteristics and three-dimensional structures was predicted using online standard softwares. The f...
متن کاملComparative conservation analysis of the human mitotic phosphoproteome
MOTIVATION A key challenge in phosphoproteomic studies is to distinguish functionally relevant phosphorylation sites from potentially 'silent' phosphorylation. Considering that relevant phosphorylation sites are expected to be better conserved during evolution than overall Serine, Threonine and Tyrosine (S/ T/ Y) residues, we asked whether this can be directly demonstrated through statistic ana...
متن کاملEvolution of characterized phosphorylation sites in budding yeast.
Phosphorylation is one of the most studied and important regulatory mechanisms that modulate protein function in eukaryotic cells. Recently, several studies have investigated the evolution of phosphorylation sites identified by high-throughput methods. These studies have revealed varying degrees of evidence for constraint and plasticity, and therefore, there is currently no consensus as to the ...
